Polymer vesicles exhibited diverse potentialapplications in catalysis, biomacromolecules and drug delivery, waterremediation, bacterial inhibition and theranostics, etc. 1-8 We recently prepared an antibacterialvesicle as an “armed” carrier of anticancer and antiepileptic drugs, showinglow cytotoxicity to normal human L02 cells but excellent antibacterialefficacy.8 Very recently, to significantlyimprove the sensitivity of a T1 MRI contrast agent and to enhance the efficacy of cancer chemotherapy, wesynthesized a noncytotoxic asymmetrical cancer targeting polymer vesicle basedon R-poly(L-glutamic acid)-block-poly(caprolactone).4 These vesicleshave a cancer-targeting outer corona and a Gd(III)-chelating anddrug-loading-enhancing inner corona, exhibiting an extremely high T1 relaxivity (42.39 mM-1s-1,8-fold better than DTPA-Gd) and anticancer drug loading efficiency (52.6% fordoxorubicin). Moreover, the DOX-loaded vesicles exhibited excellent antitumoractivity (two-fold better than free DOX). This “chelating-just-inside” strategy for synthesizing asymmetrical polymer vesicles demonstrated promisingpotential theranostic applications in magnetic resonance imaging andcancer-targeted drug delivery.
Very recently, wedesigned an EpCAM-antibody-labeled noncytotoxic polymer vesicle for cancer stemcells-targeted delivery of anticancer drug and siRNA (Figure 1).9 The EpCAMpositive CSCs-targeting vesicles showed high delivery efficacy of bothanticancer drug (DOX) and siRNA specific to the CSCs. Compared tonon-CSCs-targeting polymer vesicle, the DOX or siRNA loaded CSCs-targetingvesicle exhibited excellent CSCs killing and tumor growth inhibitioncapabilities with lower toxic to normal cells in vitro, demonstrating promisingpotential applications in nanomedicine.

简历:杜建忠,男,博士,东方学者,上海千人计划专家,上海特聘专家。现任同济大学材料学院和上海市第十人民医院教授、博导,中国生物材料学会青年委员会委员,材料学院教授委员会副主任兼秘书长,高分子材料系主任。
2004年在中国科学院化学研究所获博士学位。2004-2009年先后在谢菲尔德大学、剑桥大学从事博士后研究工作。2010年起在同济大学工作。已在JACS, Angew, ACS Nano等期刊发表70多篇学术论文。其学术论文被多家期刊引用3000多次。
