Abstract

Earlydetection and diagnosis of high-risk cancer is critical in decision-making andtailoring effective treatment. Development of new therapeutics is essential totreat the patients diagnosed with high-risk cancer and to improve theirsurvival and quality of life.  Biomolecularengineering bridges the gap between basic research and clinical needs.  Our lab is focused on translationaldevelopment of new diagnostics and therapeutics for early detection and effectivetreatment of the diseases that impair and threaten human lives. We havedesigned and developed polydisulfide-based biodegradable macromolecular MRIcontrast agents for evaluating tumor angiogenesis and targeted Gd(III) based MRIcontrast agents for detecting a cancer-related biomarker expressed in theextracellular matrix of malignant tumors.  The biodegradable macromolecular contrast agents areeffective for characterizing tumor angiogenesis and non-invasive evaluation oftumor response to cancer therapies in DCE-MRI.  The targeted contrast agents specificallybind to the biomarker in tumor extracellular matrix, resulting in strong high-resolutioncontrast enhanced molecular MRI in the tumor tissue, even small tumors withsize less than 1 mm.  We have also developedsimple and smart multifunctional delivery systems to deliver therapeutic siRNAsinto the cytoplasm of cancer cells to regulate the expression of diseaserelated genes to effectively treat life-threatening diseases, includingmetastatic and drug resistance breast cancer. The siRNA delivery systems caneffectively silence the target genes via systemic delivery to effectivelyprevent cancer progression and metastasis and re-sensitize drug resistanttumors to chemotherapy.

 

 

Biosketch

Dr. Zheng-Rong Lu isM. Frank Rudy and Margaret Domiter Rudy Professor of Biomedical Engineering atthe Department of Biomedical Engineering, Case School of Engineering, CaseWestern Reserve University.  Dr. Lu receivedhis B.S. and M.S. in Chemistry from Lanzhou University, and Ph.D. in Chemistryfrom Lanzhou Institute of Chemical Physics, Chinese Academy of Sciences atLanzhou, China.  In1992, Dr. Lu was recruited as Associate Professor of Chemistry and promoted toProfessor of Chemistry shortly after at Wuhan University in China.  In 2002, Dr. Lu was recruited as AssistantProfessor in the Department of Pharmaceutics and Pharmaceutical Chemistry atthe University of Utah.  Subsequently,Dr. Lu was promoted to a tenured Associate Professor in 2006.  Dr. Lu’s research efforts involve molecular imaging,novel MRI contrast agents, image-guided photodynamic therapy, polymeric drugdelivery systems and multifunctional delivery systems for nucleic acids.  He has over 150 peer-reviewed scientificpublications, eight book chapters and over100abstracts and four US patents.  He is aPrincipal Investigator of several major grants from the NIH.  He has served on numerous NIH study sections.Dr. Lu serves on the scientific advisory board ofPharmaceutical Research, Molecular Pharmaceutics and theAmerican Journal of Nuclear Medicine and Molecular Imaging. He is a Fellowof the American Institute for Medical and Biological Engineering.
